The Camillo Golgi Research Award-2015 in Cell Biology goes to Ponnamparambil Purushothaman Binitha and Ramadasan Kuttan from India. The research findings were published on world’s prestigious The Journal for Cell Biology. International Agency for Standards and Ratings recognizes the researchers among World's 500 Most Influential Cytologist for the year 2015 on earth. Dr. Ramadasan Kuttan is Research Director at Amala Cancer Research Centre, Amala Nagar, Thrissur, Kerala, India. He completed his B.Sc in Chemistry from University of Kerala, India. He did M.Sc in Applied Chemistry, University of Kerala, India. He also did Ph.D in Biochemistry from University of Madras, India. Binitha P. P. is Ph.D scholar at Amala cancer research centre, Amala Nagar, Thrissur, Kerala, India. He completed his B.Sc in Biochemistry from SreeKrishna College, Guruvayur, University of Calicut, India. He did M.Sc in Bio chemistry, Calicut University campus, Thenhippalam, University of Calicut, India. He also did M.phil from Avinashilingam University, India. Effect of long term oral administration (180 days) of low dose of carbofuran (5 mg/kg body weight) was evaluated on serum and different tissue enzymes of Wistar rats. The activities of mitochondrial enzymes such as isocitrate dehydrogenase (ICDH), succinate dehydrogenase (SDH), malate dehydrogenase (MDH) and NADH dehydrogenase were decreased significantly in liver homogenate of carbofuran treated animals. Hexokinase activity was found to be significantly increased in liver during chronic administration of carbofuran to rats. Glucose-6-phoshate dehydrogenase, glucose-6-phosphatase and fructose 1, 6 bisphospatase were significantly decreased in carbofuran treated rats. In addition to these there was significant inhibition of acetylcholinesterase (AchE) and marked elevation of creatine kinase (CK), lactate dehydrogenase (LDH), and gamma glutamyl transferase (γ-GT) in serum and different tissues of treated animals. The findings of this study suggested that carbofuran induced oxidative stress caused damage to rat liver mitochondrial tissues by altering mitochondrial dehydrogenases as well as glycolytic and gluconeogenic enzymes. The results also indicated that carbofuran administration produced neurological failure in Wistar rats due to inhibition of AchE and elevated the activity of CK, LDH and γ-GT in serum and tissues indicating leakage of these enzymes from damaged tissues leading to specific organ toxicity. Long term administration of carbofuran administration inhibits AchE and increased other biomarker enzymes like LDH, CK and γ-GT. The activities of citric acid cycle enzymes were found to be decreased which may be due to mitochondrial damage caused by carbofuran induced oxidative damage. Increased free radical production reduces the ability of the cell to meet the energy demands by reducing the levels of mitochondrial TCA cycle enzymes. Biochemical alterations like the increased hexokinase activity and decreased glucose-6-phosphate dehydrogenase, glucose-6-phosphatase and fructose-1, 6-bisphosphateas during chronic carbofuran administration might be due to increased metabolic need for energy requirement when alters biochemical reactions including glycolysis and gluconeogenesis. They have not included all the mitochondrial and carbohydrate metabolisms in this study. Other cellular enzymes which are affected by carbofuran should also look into. Long term carbofuran administration (5 mg/kg body weight) in rats produced significant lowering of mitochondrial enzymes. Carbofuran administration produced lowering of carbohydrate metabolism except hexokinase activity. Carbofuran administration produced lowering of AchE in serum and tissues. CK, LDH and γ-GT were found to be increased in long term carbofuran.
