In their report on Comparative study between callus, regenerated shoots and tap roots with respect to production of β-carotene, carbohydrate and vitamin C in vivo and in vitro from carrot (Daucus carota L.) they mention that Introduction: This experiment was conducted at the College of Science labs, Kufa University, on 2013 to describe methods for extraction and purification of β-carotene, carbohydrate and vitamin C content from callus tissue and tap roots mother plant (Daucus carota L.) this study aims to add further techniques in the field of plant biotechnology focusing specifically on tissue culture. Methodology: Callus induction from hypocotyls ( 2 cm pieces) by using MS medium supplemented with dichlorophenoxyacetic acid (2, 4-D) at concentration of (1 ,2, 3) mg/l. Shoots regenerated from callus by using MS medium supplemented with ( IAA )at concentration of (1) mg/l and (BA) benzyl adenine at concentration of (0.2) mg/l in dark condition ,fresh standard solution of control(Ascorbic acid, Glucose and carotenoids).The content of β-carotene was done by using HPLC (high performance liquid chromatography), vitamin C and carbohydrate were done by using Spectrophotometric method at 620 nm and compare of the quantitatively of β-carotene, carbohydrate and vitamin C with these content in tap roots of mother plant, Shoots regenerated and callus tissue. Results: This study include alcohol extraction by using acetone .The result revealed that, the superiority of β-carotene, carbohydrate and vitamin C in Callus content and regenerated shoots than the content in tap roots mother plant and increase the content of these compound in the shoots of regenerated plant with the content in callus tissue. Under their leadership the research area is surely likely to expand beyond the horizons. It will be worthy to explore the further potential under their legacy. Kareem T.
